Age related cataract is leading cause of blindness, affect 46% people
worldwide (Asbell PA et al., 2005). Surgery is the only effective treatment for cataract and the exact
mechanism for its formation is still not very clear. Although cataract surgery is
recognized as being one of the safest operations, there are significant rate of
complications, leading to irreversibly blind eyes (Manikandan R et
al., 2009). The cataract formation
in the eye is generally associated with several risk factors including ageing, oxidative
stress, and impaired lifestyle such as the use of tobacco, abuse of alcohol and
unhealthy diet which accelerate the ageing process (Kang L et al., 2015; Kubota M  et al., 2016; Goutham G et al., 2017). Previous
studies reported that metabolites of naphthalene cause oxidative stress that
contributes to lens opacity, it is also found in our study that chronic
administration of naphthalene to the animals of control group produced
significant cataractogenic effect (Pandya U  et al., 2000). It enhanced the oxidative stress in lens
as indicated by reducing antioxidants (CAT, SOD, GPX, and GSH) and elevating
lens MDA level. The antioxidant level is determitant of oxidative stress
markers. Oxidative stress is a common initiator of many
age-related conditions such as atherosclerosis, Alzheimer’s disease and some
diseases of the eye like macular degeneration, glaucoma and cataract (Geraldine P et al., 2006). Excessive
free reactive oxygen species (ROS) generation leading to oxidative stress,
wherein antioxidant defense mechanism is disrupted or depletion of antioxidants
or both (Spector A, 1995). The
elevation of ROS formation further leads to oxidative damage on cellular
protein and lipids (Pandey KB and Rizvi
SI, 2010), which were observed
in control group/our study.

Resveratrol, is a silent information regulator type
1(SIRT1) activartor found naturally in many plants and herbs, has significant
antioxidant activity against free radicals both in vitro and in vivo (Reference for in vitro and in vivo). It
is established that SIRT1 is associated with several age-related diseases, such
as age related macular degeneration,
metabolic diseases, cardiovascular disease and neurodegenerative diseases (Bhattacharya et al., 2012; Zeng et al.,
2009).

Silent
mating type information regulation 2 homolog 1 (SIRT1) is an NAD-dependent class
III histone deacetylase that plays crucial roles in multiple biological
processes, including the oxidative stress-induced apoptosis, lipid metabolism,
ageing process, many age-related diseases including cardiovascular diseases and
neurodegenerative pathologies (Marmorstein
R, 2004, Michan S et al., 2007, Longo VD et al., 2007, Canto C et al., 2012).
SIRT1 is localized in the nucleus and cytoplasm of most ocular tissues
including the cornea, lens, iris, ciliary body, and retina (Mimura T et al., 2013). The upregulation of SIRT1 protects against
various ocular diseases such as retinal degeneration, optic neuritis, and
uveitis (Mimura T et al., 2013).
However, there are few known facts about the relationship between SIRT1 and
cataract, and it is unknown whether SIRT1 protect age related cataract against
oxidative damage and imbalance of calcium homeostasis, but there were some
clues that suggest this possibility (R).

Resveratrol
was reported to be an activator of SIRT1 both in vivo and in vitro (Borra et al., 2005; Howitz et al., 2003).
Resveratrol, a type of natural phenol known as a stilbenoid was found to prevent
oxidative stress-induced retinal diseases such as AMD and oxidative stress in
mouse lung, liver and kidney (Wu et al.,
2007, Sehirli O et al., 2008). It has been shown to upregulate SIRT1 and its substrate as a
structural facor and by increasing SIRT1 mRNA expressions (Borra  et al., 2005, Zhao K et al., 2004, Bai L et
al., 2008).

The results of the present study have shown
that chronic administration of RSV at the dose of 40 mg/kg/day, p.o
concurrently with naphthalene significantly abolished the oxidative stress
effects of naphthalene administration.
